In the fructose syrup production plant the isomerization converts glucose which is not very sweet to fructose, the most sweet of the natural sugars. Syrups from this process compete with sucrose (cane sugar) in many food applications. Almost all manufacturers of soft drinks use high fructose syrups because they are less expensive than sucrose. This was devastating to world prices of cane sugar and crippled the economies of some countries.

The isomerization of glucose to fructose in the fructose syrup production plant is part of the glycolysis cycle that converts glucose to pyruvate. The way this is done is to isomerize the aldehyde (hemiacetal) glucose to the ketone (as a hemiacetal) fructose,and make another phosphate ester. The isomerization takes advantage of the ease of breakage of a C-H bond which involves a carbon next to a carbonyl carbon. This is important in the next step which cleaves the bond between carbons three and four of fructose. It is noted that this bond involves the carbon next to the carbonyl carbon of fructose. This cleavage would not have been possible without the isomerization of glucose to fructose, because the carbonyl group of glucose is too far from carbons three and four to make that bond breakable.

Glucose isomerase (D-glucose ketoisomerase) causes the isomerization of glucose to fructose. Glucose has 70-75% the sweetening strength of beet sugar (sucrose), but fructose is twice as sweet as sucrose. Thus, processes for the manufacture of fructose are of considerable value.

Novo Industries has developed glucose isomerase from B. coagulans for commercial use. In this immobilized enzyme process, the microorganism carries out a direct isomerization of the glucose. This glucose isomerase is primarily a xylose isomerase, so xylose, or a xylose-containing compound must be added for the induction of the enzyme.

How to produce glucose syrup from starch ?

Glucose syrup, also known as candy glucose, is a kind sweet syrup made from the hydrolysis of starch. Maize /corn  is commonly used as the source of the starch ,so the glucose syrup is called "corn syrup" . But glucose syrup is also made from other kind starch, such as cassava starch , potato starch ,wheat starch and so on .

Then , how to produce glucose syrup from starch in glucose syrup processing plant ?

The production process of glucose syrup from  starch in modern glucose syrup processing plant is as below : 

(1) Starch milk adjustment
Starch milk coming from starch plant is adjusted to DS28-32% and proper pH and temperature. Or mix the dry starch with water to reach above condition for next section.

(2) Liquefaction
Starch milk is sent to buffer tank then liquefied by jet cooker with the aid of enzyme.

(3) Saccharification
The liquefied material is sent to enzyme saccharification tank for reaction. After being kept in holding tube the required DE value of the glucose will be reached. Then the material is sent to flash for cooling then to the neutralization tank for suitable pH value.

(4) Filtration and decoloring
Glucose is cleaned by removing the protein and other foreign items in rotary vacuum filter and filter press. Active carbon is needed for decoloring at proper temperature.

(5) Ion exchange
Tiny foreign items and bad odor is removed via ion exchanger. This equipment is used for high quality product.

(6) Evaporation
The glucose is totally cleaned through safety filter then sent to evaporator for concentrating to reach the required DS as final product.


In the world , all the corn glucose syrup processing plant use  corn to make corn starch first  , then use the corn starch to produce the glucose syrup .
